Sigmoid curves, non-linear double-reciprocal plots and allosterism.

چکیده
1. The theory of plane curves was applied to the graphical methods used in enzyme kinetics and a mathematical analysis of the possible graph shapes is given. 2. The belief that allosterism can be inferred from steady-state data alone is subjected to criticism and the mathematical significance of sigmoid curves and non-linear double-reciprocal plots is explored. 3. It is suggested that the usual methods of interpreting steady-state kinetic data are often based on over-restrictive assumptions which prevent maximum utilization of the available data. 4. Methods for obtaining the degree of the rate equation from graph shapes obtained directly from initial-rate measurements and from replots of asymptotic behaviour as chi approach the level 0 and chi approach the level infinity are discussed. 5. Detailed proofs of the theorems given in the text have been deposited as Supplementary Publication SUP 50049 (10 pages) at the British Library (Lending Division), Boston Spa, West Yorkshire LS23 7BQ, U.K., from whom copies can be obtained on the terms indicated in Biochem. J. (1975), 145, 5.

Enzyme Concentration Influences on Activity Measurements
Moat plots of enzyme concentration versus catalytic activity are linear, or fall on a single arm of an hyperbola, allowing treatment in the double reciprocal manner of "maximum rate of product formation" kinetics. Since curves of similar, if undefined, shape arise from many experimental measurements, the double reciprocal approach with its characterizing constants, can be employed to give an ap...
متن کاملThe inhibition of skeletal-muscle fructose 1,6-diphosphatase by adenosine monophosphate.
1. The present study extends the finding of Krebs & Woodford (1965) that muscle fructose diphosphatase is more sensitive to AMP inhibition than liver fructose diphosphatase. 2. Hen breast fructose diphosphatase has a K(i) for AMP of 0.1mum; the plot of percentage inhibition is non-sigmoid and the reciprocal plot of activity against AMP concentration is sometimes linear. 3. Percentage inhibition...
متن کاملAllosteric behavior in transducin activation mediated by rhodopsin. Initial rate analysis of guanine nucleotide exchange.
Photolyzed rhodopsin acts in a catalytic manner to mediate the exchange of GTP for GDP bound to transducin. We have analyzed the steady-state kinetics of this activation process in order to determine the molecular mechanism of interactions between rhodopsin, transducin, and guanine nucleotides. Initial velocities (Vo) of the exchange reaction catalyzed by rhodopsin were measured for various tra...
متن کاملStudies on the kinetic mechanism of oxidative phosphorylation.
The kinetics of the synthesis of ATP from ADP and Pi by beef heart submitochondrial particles were examined. When Pi was the variable substrate positive cooperativity was observed, whereas if ADP was varied, linear double reciprocal plots were obtained. The analog of Pi, thiophosphate, was a noncompetitive inhibitor of ATP synthesis with respect to ADP, while the analog of ADP, AMP (CH2)P, was ...
متن کاملOxidative phosphorylation: a new biological function for lipoic acid.
In experiments similar to that of Fig. 1, linear plots of 1/K+ influx against the reciprocal of the external K+ concentration were fitted to the data by the method of least squares. Values of K, and VmaX. were calculated from these plots according to the usual relationships: 1/Vmax. equals 1/K+ influx when l/[K+] equals zero, and -1/K, equals l/[K+] when 1/K+ influx equals zero. The values of K...
